چکیده

HIS investigation of morphological mutants in Neurospora crassa was underTtaken to examine a wide spectrum of such strains and to obtain information regarding their number and distribution in each of the seven linkage groups. This information, as well as a comparative study of their morphological characteristics, is important both theoretically and practically in facilitating further investigations of the biochemical defects involved. The present report describes the linkage to known markers of 90 morphological strains of independent origin. Of these, 58 have designated as new loci; the remainder appear to represent alleles either of the new or previously described loci. The order of the genes has been determined only in group V (MORGAN, GARNJOBST and TATUM 1967). The differences between strains in type of mycelial growth, both from the ascospore and upon transfer, and the distinct differences between some strains in hyphal morphology have suggested a new and practical classification of the morphological mutants. Six classes are defined, illustrated, and discussed.

Biotransformation of Hydrocortisone by Neurospora crassa

The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...

Characterization of the Temperature-Sensitive Mutations un-7 and png-1 in Neurospora crassa

The model filamentous fungus Neurospora crassa has been studied for over fifty years and many temperature-sensitive mutants have been generated. While most of these have been mapped genetically, many remain anonymous. The mutation in the N. crassa temperature-sensitive lethal mutant un-7 was identified by a complementation based approach as being in the open reading frame designated NCU00651 on...

Isolation and characterization of low-kynureninase mutants of Neurospora crassa.

Two kynureninase activities are known in Neurospora crassa, one of which (kynureninase I) is inducible, the other (kynureninase II) being constitutive. A method is described for the isolation of low-kynureninase mutants of N. crassa. When grown on an inducer, the mutants show significantly less kynureninase I activity compared with wild type, whereas constitutive kynureninase II activity is una...

A Cytoplasmic Character in Neurospora Crassa

Nuclear fractions isolated from mutants of Neurospora produced no effect when microinjected into mutants with complementary biochemical requirements. DNA isolated from the nuclear fractions similarly injected also had no effect. Mitochondrial fractions isolated from an abnormal inositolless strain (abn-1) produced drastic changes in the rate of growth, morphology, reproductive characteristics, ...

Complementation analysis of linked circadian clock mutants of Neurospora crassa.

A fourth mutant of Neurospora crassa, designated frq-4, has been isolated in which the period length of the circadian conidiation rhythm is shortened to 19. +/- 0.3 hours. This mutant is tightly linked to the three previously isolated frq mutants, and all four map to the right arm of linkage group VII about 10 map units from the centromere. Complementation tests suggest, but do not prove, that ...
